In the spring of 2015, reports of Nystatin being used for PGAD and on the Procepia help forum were noted on the PSSD forum. In may of that year, 2 reports of Nystatin helping PSSD were reported by user “Continue”.

man… me and a friend of mine got improvements with nystatin + diflucan for PSSD.

nystatin gave me dreaming + morning woods, and diflucan gave me spontaenous erections, better erections and sensations+orgasm back.

Another Procepia recovery story reignited the conversation around Nystatin in the fall of 2016. “Continue” confirmed that the friend (BRMAN from PSSD forum) he has spoken of earlier had now made a recovery.

Yes, a friend of mine (who was, in the past, an active member in several PSSD foruns, including this one) was fully healed with Nystatin, but that was the latest part of his healing.

In November of 2016, forum user sulawesi reported:

I’m taking 7pills of Nystatin three times a day. But I think I will change to 2 doses at 10 pills a day. I know that’s a lot and a little crazy… but I think need those high doses to get any results.

After 3 weeks of treatment he said:

Guys, I really have to say that right now I feel 98.7% back to normal!!!

It is currently unknown whether these success stories from nystatin are placebo and luck, or if Nystatin is a genuine treatment option for PSSD. Further research and experimentation are needed before further conclusions can be made.

The relative number of success stories from the small number of testers leads me to believe that Nystatin’s ability to help PSSD* in some suffers is “Likely”.

Pharmacology

Nystatin is a prescription anti-fungal in the United States, but is over-the-counter in most parts of the world.

Why would we suspect an anti-fungal to be a candidate for PSSD treatment?

Initially, we wouldn’t. In fact, this is why Nystatin was first reported as helpful for PSSD as an accident. When we look deeper, however, we see that Nystatin modulates activity of the serotonin system. This is another line of evidence for the 5-HT1A model of PSSD.

The sterol-binding antibiotic nystatin differentially modulates ligand binding of the bovine hippocampal serotonin1A receptor.

“Nystatin considerably inhibits the specific binding of the antagonist to 5-HT1A receptors in a concentration-dependent manner.”

It’s also possible that Nystatin is giving benefits through its action on Nitric Oxide

Nystatin-induced nitric oxide production in mouse macrophage-like cell line RAW264.7.

“Surprisingly, treatment with nystatin induced only NO production and iNOS expression in RAW264.7 cells. At the concentration used, no changes in the expression of GM1 ganglioside, a lipid raft marker on RAW264.7 cells, was seen.”

 “The results suggest that a moderate concentration of nystatin induces NO production by Na-pump activation through the PI3 kinase/Akt/NF-kappaB pathway without affecting the condition of lipid rafts.”

There is a distinct void of information regarding Nystatin and its serotonergic activity. Future research would need to be conducted in order to gain a better understanding of why exactly Nystatin has helped in PSSD cases.
